 Antibody Specificity

Based on immunoelectrophoresis and/or ELISA, the antibody reacts with whole molecule mouse IgG. It also reacts with the light chains of other mouse immunoglobulins. No antibody was detected against non- immunoglobulin serum proteins. The antibody may cross-react with from other species.

如何选择二抗

通常情况下，某一实验中可能同时有几种二抗可供选择，如何能选择到最适合该实验的二抗，需要综合以下几个方面进行考虑：

一、一抗的种属来源

一抗是什么物种来源的，相应的二抗也要是抗该物种的抗体。

二、一抗的免疫球蛋白亚型

二抗需与一抗的类别或亚型相匹配。例如一抗是IgM，则须选择抗IgM的二抗。如果一抗是某一亚型(如IgG1、IgG2a、IgG2b、IgG3、IgG4等)，则可选择能与所有亚型结合的抗IgG二抗，也可选择专门针对该亚型的二抗。

三、实验方案

根据实验，选择酶标记(如HRP)的二抗，还是荧光素标记的二抗。根据仪器的激发光和检测通道，选择合适的荧光素。

四、选择哪种形式的二抗，是整个分子还是F[ab’]2片段？ 整个IgG分子：此种抗体适用于多数情况。

F[ab’]2 片段：该抗体由 2 个二硫键连接的 Fab 片段组成，用于特定的情况中，如需要避免抗体与具有

Fc 受体的细胞结合，降低非特异性背景。
